I have the following setup in my hosting account

    _parent
    |_domain.com
    |_wp (wordpress 3.0.1 install)

Wp-hive is installed (and active) in the wp node

    In my hosting account:
    http://domain.com is pointed at parent/domain.com
    http://blog.domain.com is pointed at parent/wp

In Wp-hive admin I have set up:
    Domain name: blog.domain.com
    Sub Directory: /
    Prefix: dvs_

domain.com resolves exactly as I want.
    blog.domain.com does not resolve

Update: In FF it is giving me this error:

    The page isn't redirecting properly

Firefox has detected that the server is redirecting the request for this address in a way that will never complete.

    * This problem can sometimes be caused by disabling or refusing to accept
    cookies.

Check the 'home' and 'siteurl' options. They should both be set to the same thing, and sometimes the trailing slash (either present or not present) can cause redirect issues.

Also check that .htaccess is up to date. Try deleting / renaming .htaccess and see if you regain access to the site. If so, re-generate it by saving the permalinks.

From what I can tell from your setup domain.com is treated independently of blog.domain.com, so I don't see any reason for the conflict.
